Llama and alpaca comparative sperm head morphometric analysis

چکیده
South American camelids are classified in four different species included in two genera. Llama (Lama glama L.) and alpaca (Vicunya pacos, L.) are the domestic species, presenting a high economical interest. This classification is based on historical, evolutionary and morphological criteria, but the level of hybridization and the total reproductive compatibility make it difficult to use the “biological criteria” based on reproductive isolation. The aim of the work was to see if there are differences on spermatozoa morphometry indicating a pre-zygotic isolation mechanism based on sperm competition. Nine adult Lanuda (Ch’aku) llamas and eleven Huacaya alpacas were used. After obtaining the samples using an artificial vagina, semen smears were stained with Hemacolor kit. Samples were automatically analysed using the morphometry module of the ISASv1 CASA system. Almost 100 sperm cells were analysed per sample. The following parameters were calculated: Length, Width; Perimeter, Area, Ellipticity, Rugosity, Regularity and Shape Factor. Most of the parameters (except rugosity and regularity) were significantly different between both species, being greater in alpaca than in llama. Looking to the variation data, both intra and inter-animal coefficients of variation (CV) were also different, but in this case the variation was higher in both CV in llama than in alpaca. From the point of view of the sperm competition, all these data would suggest that both species are in the process of a gamete isolation process. A good selection of sperm characteristics could help farmers to define the “purity” of each animal using the morphometrical analysis, not much more effective than other techniques, but much simpler and less expensive.

Comparison of three different staining methods for the morphometric characterization of Alpaca (Vicugna pacos) sperm, using ISAS® CASA-Morph system
Sperm morphometry is a part of the semen analysis based on CASA technology and has shown a big role in the prediction of male fertility. This analysis implies the use of stain techniques, although it has been shown that staining can make dramatic changes in the cell morphometry in different species. The aim of the present work was to evaluate the differences in sperm morphometry in Alpaca, intr...

The effect of four enzymes: collagenase, fibrinolysin, hyalurodinase, and trypsin were recorded on the viscosity, motility, percent live spermatozoa and acrosome integrity of Llama and Alpaca semen. Semen samples were collected using a modified artificial vagina for each of the five llamas and five alpacas. A 25% solution of the of enzyme at a concentration of 1mg/ml was added to the ejaculate....
متن کاملGenetic analysis reveals the wild ancestors of the llama and the alpaca.
The origins of South America's domestic alpaca and llama remain controversial due to hybridization, near extirpation during the Spanish conquest and difficulties in archaeological interpretation. Traditionally, the ancestry of both forms is attributed to the guanaco, while the vicuña is assumed never to have been domesticated. Recent research has, however, linked the alpaca to the vicuña, datin...
متن کاملSpermiogram and sperm head morphometry assessed by multivariate cluster analysis results during adolescence (12-18 years) and the effect of varicocele
This work evaluates sperm head morphometric characteristics in adolescents from 12 to 18 years of age, and the effect of varicocele. Volunteers between 150 and 224 months of age (mean 191, n = 87), who had reached oigarche by 12 years old, were recruited in the area of Barranquilla, Colombia. Morphometric analysis of sperm heads was performed with principal component (PC) and discriminant analy...

The aim of this study was to compare the sperm nuclear and acrosomal morphometry of three species of domestic artiodactyls; cattle (Bos taurus), sheep (Ovis aries), and pigs (Sus scrofa). Semen smears of twenty ejaculates from each species were fixed and labeled with a propidium iodide-Pisum sativum agglutinin (PI/PSA) combination. Digital images of the sperm nucleus, acrosome, and whole sperm ...
